Gut Microbiome Composition Is Associated With Future Onset of Crohn’s Disease in Healthy First-Degree Relatives
Raygoza Garay et al (including Platform 3 Lead Dr. Karen Madsen and CMI2 Team Grant Leads Dr. Anne Griffiths, Dr. David Mack, Dr. Michael Surette, & Dr. Bruce Vallance). Gastroenterology. 30 May 2023.
Highlights: In this prospective cohort study, researchers enrolled 3,483 healthy first-degree relatives (FDRs) of patients with Crohn’s disease (CD), and monitored them for a median of 5.4 years, to test if their gut microbiome composition could be used to predict the risk of developing CD. In this cohort, 73 individuals developed CD during the study. They used a validation cohort to develop a microbiome risk score (MRS) model, which was able to identify individuals that developed CD up to 5 years before disease onset. This is the first study to show that gut microbiome composition is associated with future onset of CD.

Birth Mode Does Not Determine the Presence of Shared Bacterial Strains between the Maternal Vaginal Microbiome and the Infant Stool Microbiome
Dos Santos et al. (including IMPACTT Co-Applicant Dr. Janet Hill). Microbiology Spectrum. 20 Jun 2023.
Highlights: Previous studies have found links between delivery mode (vaginal vs. c-section) and a variety of diseases, such as asthma, type I diabetes, and other chronic and autoimmune diseases. It has been hypothesized that early life gut microbiome dysbiosis may play a role in the development of these diseases, especially since c-section infants are not immediately exposed to the maternal vaginal microbiome at delivery. In this study, the researchers set out to determine if the maternal vaginal microbiome is vertically transmitted to infants. They sequenced 585 maternal vaginal microbiomes and 1,027 infant stool microbiomes (collected at multiple time points), and found that 204/585 mother-infant pairs showed at least one potential ASV transfer event. Delivery mode did not affect the distribution of these potential transfer events, nor did antibiotic exposure. Based on these results, the researchers show that vertical transmission of the maternal vaginal microbiota is likely limited and that transmission from other maternal body sites, including the gut and breast milk, may compensate for the lack of maternal vaginal microbiome exposure during c-section delivery.
