The rise to power of the microbiome: power and sample size calculation for microbiome studies

Highlights: In this review, the authors summarize statistical approaches to calculate sample sizes for typical microbiome study scenarios, including those that hypothesize microbiome features to be the outcome, the exposure, or the mediator. This review is intended to be a resource to facilitate sample size calculations that are based on testable hypotheses across several dimensions of the microbiome, and includes relevant R scrips to conduct some of these calculations.

New Publications in the Microbiome World

Biogeographic variation and functional pathways of the gut microbiota in celiac disease
Constante et al. (including CMI2 Team Grant Lead Dr. Michael Surette). Gastroenterology. 8 Jul 2022.
Highlights: Altered gut microbiota has been implicated as a risk factor for celiac disease. In this study, researchers characterized the microbiota along the GI tract of celiac disease patients and gnotobiotic mice using Illumina sequencing and predicted gene function. They determined luminal and mucosal microbial niches along the gut in celiac disease patients, and identified novel microbial proteolytic pathways involved in gluten detoxification that are impaired in celiac disease.

Altered gut microbiome composition and function are associated with gut barrier dysfunction in healthy relatives of patients with Crohn’s disease
Leibovitzh et al. (including Platform 3 Lead Dr. Karen Madsen and CMI2 Team Grant Leads Dr. Anne Griffiths & Dr. David Mack). Gastroenterology. 16 Jul 2022.
Highlights: In this study, researchers aimed to assess if alterations in gut microbial composition and functional pathways were associated with gut barrier function, using a cohort of healthy first-degree relatives of patients with Crohn’s disease. They found that subjects with impaired gut barrier function had reduced alpha-diversity and altered beta-diversity as compared to subjects with normal gut barrier function. Impaired gut barrier function was also associated with eight genera and 52 microbial pathways, suggesting that the gut microbiome community and pathways are associated with changes in gut barrier function.

Childhood body mass index and associations with infant gut metabolites and secretory IgA: findings from a prospective cohort study

Highlights: In this prospective cohort study, researchers set out to determine associations between infant fecal gut metabolites and sIgA (measured at 3-4 months) and BMI z score (calculated at 1 and 3 years). They found that high fecal formate in infancy was associated with significantly lower BMI x score, while high butyrate was associated with higher BMI z score (at the 3 year mark only). No associations were seen between sIgA and BMI z score.
