Linking microbial genes to plasma and stool metabolites uncovers host-microbial interactions underlying ulcerative colitis disease course
Schirmer et al. (including CMI2 Team Grant Leads Dr. Anne Griffiths & Dr. David Mack). Cell Host & Microbe. 11 Jan 2024.
Highlights: In this study, the researchers combined paired metagenomics, metabolomics (stool and plasma), and culturomics in order to link disease-associated microbes to microbial metabolites in a pediatric inception cohort focused on ulcerative colitis (UC). They identified host-microbial interactions correlated with disease activity, inflammation, and the clinical course of UC in this cohort. In particular, they found metabolite changes in the stool in severe disease, including increased dipeptides and tauro-conjugated bile acids, as well as decreased amino-acid-conjugated bile acids. They also identified increased plasma polyamines with increasing disease severity. They further discovered that UC-associated Veillonella parvula was able to metabolize immunosuppressive thiopurine drugs, potentially impairing the therapeutic response.
