New Publications in the Microbiome World

Highlights: In this study, the researchers used data from their recent randomized placebo-control phase 2 clinical trial in adults with obesity and metabolic syndrome. Participants were grouped into HOMA-IR responders and non-responders (HOMA-IR is an assessment for insulin resistance). They found that baseline fecal microbiota composition of the recipients could predict insulin resistance improvements at week 6 of the trial, with the genus Prevotella being identified as an important predictor (lower relative abundance being associated with the responder group). Responders also had increased bacterial richness after FMT and a more consistent engraftment of donor-specific bacteria. Microbial engraftment was also strongly associated with recipient factors at baseline such as initial gut microbial diversity, fiber and nutrient intakes, inflammatory markers, and bile acid derivative levels. The findings of this study provide a basis for the development of precision microbial therapeutics for the treatment of metabolic syndrome.

Highlights: One of the challenges of creating synthetic microbiomes is obtaining a representative bacteriophage population during the bacterial isolation process. In this study, the researchers show that dilution-based isolation methods generate synthetic gut microbiomes that are heavily depleted, if not devoid, of virulent viruses. When they re-introduced a virulent dsDNA virome to a 73-strain synthetic gut microbiome in a bioreactor model of the human colon, the researchers found that these virulent viruses targeted susceptible strains without significantly altering the overall community structure or metabolism. They conclude that such viruses, if reintroduced to synthetic gut microbiome models, can have a targeted effect on community assembly, metabolism, and prophage replication.
